Biocatalysis - immobilized enzyme biotransformations

Position summary:
The laboratory of Professor Richard A. Gross at NYU-Poly (Brooklyn, NY) is performing fundamental, exploratory and applied research to address some of our planet’s most challenging problems. In 2008, Professor Gross founded SyntheZyme that is commercializing technology developed by his laboratory. The Gross group is developing new knowledge and technologies in biocatalytic and chemo-enzymatic routes to biobased materials including monomers, polymers, surfactants and other biochemicals. The Gross research group is currently seeking a postdoctoral fellow to work on cell-free enzyme catalyzed biotransformations.

Job Description

We seek a person skilled in studies of protein immobilization, biocatalytic transformations using immobilized enzymes and engineering aspects for immobilized reactor design. The primary enzyme families to be used in this work are lipases, esterases and proteases. Enzyme purification may be required prior to immobilization. Key issues to address will be optimizing enzyme activity on immobilization supports, assessing immobilized catalyst recyclability and associated problems that may occur due to protein deactivation, enzyme leaching and physical deterioration of matrix materials upon re-use. The successful candidate should be experienced in common methods for enzyme activity assays, purification and determination of purity. The successful candidate should also be experienced in methods for studying biocatalytic transformations in various reactor configurations including batch, packed column and milli-reactor systems. Furthermore, the successful candidate can demonstrate their experience and skills through scholarly publications. This position will be filled within 90 days of posting so that the candidate should be available to start at SyntheZyme during that time period.

This work will be performed as a joint project with a client company. The successful candidate will have the opportunity to transition from working at SyntheZyme to the client company.

The successful candidate will be located at the NYU-POLY campus where SyntheZyme currently occupies laboratory and office space.

The successful candidate will have the opportunity to publish their work as well as be part of a large team within the Gross group at NYU-POLY that consist of undergraduate, master students, Ph.D. and postdoctoral fellows.

In summary, the successful candidate will possess the following characteristics:

• Plan, execute, and analyze enzyme characterization, immobilization and biotransformation experiments.
• Apply the latest knowledge in methods for enzyme immobilization and characterization of immobilize enzyme systems.
• Develop biocatalytic processes using immobilized proteins in macroporous matrices, gels and milli-channel reactor configurations.
• Use common methods to follow enzyme reactions such as NMR, GC-MS and LC-MS.
• Provide feedback and collaborate with analytical chemists and application scientists at SyntheZyme, NYU-POLY and the client company.
• Use scientific literature to address questions of enzyme stability, characterization, purification, immobilization as well as analysis of products formed during enzyme transformations.

Requirements

• Ph.D. in Biochemical Engineering, Chemistry, Chemical Engineering, Applied Biochemistry, Biotechnology or a related discipline.

• Hands-on experience in most of the following skills is highly desired: protein purification, assess protein purity, monitor protein adsorption kinetics, assessing immobilized enzyme recyclability, optimizing enzyme activity by varying conditions during immobilization, selecting immobilization supports, use of various immobilized reactor systems such as batch, packed column and milli-reactor systems, strong familiarity with product analysis by common structural analysis methods, knowledge of process engineering for scale-up.

• Due to the highly collaborative nature of the position, excellent interpersonal and communications skills are an essential characteristic of a successful candidate.

• Able to establish and maintain effective working relationships while collaborating with other scientists, engineers, and technicians in closely related disciplines.

• Strong organizational, problem solving, and time-management skills are essential
